Research about the effectiveness of consortia bacteriaindigent in reducing heavy mercury (Hg) as the solution of liquid waste of flour industry has done after discovering that the liquid waste of flour industry containing up to 1,1093 mg/L of Mercury. This is a violation of Government regulationPP No.82 Th. 2001, which state that the minimum limit of Mercury on liquid waste approximately 0,001 mg/L at most. This is because Mercury is a poisonous heavy metallic element that may harm the ecosystem and creatures around it. One solution toward this problem is by using indigent redactor bacteria such as Pseudomonas stutzeriandActinomyces viscosus. Since the research on the bio reduction process is still needed, it is important to develop handout for biotechnology learning material so that the university students can yield some more information related to reduction process of heavy mercury by using indigent bacteria. The purpose of this research is to verify the effectiveness of indigent bacteria consortia in reducing heavy mercury (Hg), afterward the result of the research would be developed into handout for university students for further research.

This is a research and developmentresearch which is established in two phases. First phase is a laboratory experimental research and at the second phase,the researcherdevelop the handout for Biotechnology course on sub-chapter environmental biotechnology at State University of Malang. The reduction test done by inoculating isolate bacteria using several concentration variation: 3%, 4%, 5%, 6%, and 7% on 100ml liquid waste. The isolate bacteria used was A:P stutzeri, B: A. viscosusandconsortium AB: which is mixture ofP. stutzeriandA. viscosus. The result of the Hg reduction analyzed by using ANAVA statistic test. After that, the researcher test the liquid waste after the trials by using consortium AB with 7% concentration toward the survival rate of goldfish (Carracius auratus). The goldfish kept at different waste’ concentration, which is 25%, 50%, 75%, and 100% of 6 L of total media.Second phase of the research is the handout developing based on Borg dan Gall(1983) until the 7th phase. The handout, afterward, implemented to the Biology students (6th semester of year 2013 students of Offering B). The research design is One Group Pretest and posttest data were analyzed using the formula gain score in addition to the results of the pretest and posttest evaluation of student work on the problems that are measured using the lattice evaluation questions. Affective learning outcomes were measured using questionnaires affective, and science skills are measured using a rubric science skills.

The results of experimental research data showed that bacterial consortia indigen AB concentration of 7% as effective Hg reduction was 74.5%. Consortia of bacteria gives better results than the use of single isolates, selected for their synergistic interaction in the production of enzymes that can optimize work processes in reducing contaminants. Data bioassay results show that all the fish are not able to survive for seven days of observation. Fish deaths occurred because the content of Hg is still above the minimum quality standards set by the government. In addition to fish kills also occur due to the synergistic phenomenon, which is a combination of two or more substances contained in the environment that can strengthen toxic to fish. Media instructional handouts as supporting learning resources to improve learning outcomes shown on the increase in the average value of 39.60 into 83.30 classical pretest on posttest, so that the values obtained with a 0.71 gain score high for the interpretation of the classical value of 86.5 with criteria nilaihuruf A-. Affective learning results saw strong increase in the first meeting, second, and third respectively acquire classical values 82.9, 85.7, and 90.8 with A- criteria, the value of the report in the classical scored 80.5 with the letter B + value criteria. The existence of a learning resource handout equipped with practical activities to help students understand the material more easily, science skills acquired value of 83.7 with a value criterion B +
